I'm not sure what you really mean, I have heard it defined two ways.

Sister cities overseas, is usually symbolic for the most part, for small towns. It may that the founders of one city link up with the town where they immigrated from, or where their ancestors are from. Or it may be two cities in different countries that have a similar way of life based on the lay of the land or the type of industry found there. You may see a visit or an exchange now and again, for a parade or whatever, but it is largely in rememberance of a town's heritage or a gesture of diplomacy.

A similar term is twin cities, and may be referred to as sister cities since THE Twin Cities are in Minnesota (Minneapolis-St Paul). These are smallish cities that are close in size and close together on the map, and they kind of function together to meet the needs of their citizens. In one town, for example, there may be a lot of industry, where in the other, there is a big college or hospital. They rely on each other economically, for jobs and shopping and tourism, but they have their own separate city limits, and their own governments, utilities, and schools. They are different from major metros in that there is no larger city hubbing all the smaller suburbs together.

Both are fairly obscure terms. Hope this helps.

The city I live in has sister cities in Argentina, China, Ecuador, France, Germany, Ghana, Russia, and the United Kingdom.

There are regular cultural exchanges, and recently members of a gymnastic club visited from the sister city in France.

I think exchanges such as this helps us understand those from different backgrounds and realize how similar we are with each other no matter which country we are from.

They create great photo ops for the politicians who set them up, but I have never seen one instance where they benefited the citizens of the sister cities.
