Stay at the flamingo. It is right in the center of the strip, and has a monorail station attached to it. Very convenient, and fairly inexpensive. Expect to do a lot of walking in Vegas, so being central is good, and the monorail is smooth, and a great way to get around. There are also monorail stops at other hotels on the strip. If you are going to drink a lot I would recommend buying alcohol, and having a couple drinks in your room first....much cheaper. Or you can get free drinks while gambling...but they take a long time. Club drinks are expensive. Oh and Front-of-Line (or VIP passes) are worth it at the clubs. Here is a great las Vegas website, they seem to always have the hot new thing listed.

Stay away from package deals ( air & hotel ) Cheaper to book seperately. NYNY is a great place for first timers. Great location and a younger crowd stay there. The Hard Rock is great for the younger crowd but it isn't the easiest to get to. I do not recommend renting a car in Vegas. Driving is horrible, not for the timid. With a strip hotel you have so much at walking distance. Stay away from Circus Circus ( kids ), Sahara and Statosphere ( too far away ) and the Riviera ( oldest on the strip ). Do not even think about downtown LV. The crowd their is really old.
