The reason that brassiere is singular is because the person is talking about one of them. The reason that panties is plural is because the person is then talking about more than one pair.

I think its more likely because panties are generally harder to get off a woman, both literally and figuratively, so the extra effort makes it seem to a guy like he has to get off more than one (hence, plural). But getting a chick out of her bra takes almost no effort at all, assuming she is even wearing one, so nothing there is worth the respect of plurality.
Hahaha! Just kidding! I dunno why. My best guess is because bra comes from brassierre (a singular word) and panties from pants (a plural word).

because the bra goes around them once and you have to put 2 legs through the panties
Therefore one is singular and one is plural
