Looking at car hire, but what is the road conditions like at this time of the year?

The road conditions are fine. After October, there can be snow/ice conditions.

The best route from NYC:

I-80 to I-380 (metro NJ area to Scranton, PA)
I-380 to I-81 North (Scranton PA to Syracuse)
I-81 to I-690 (Syracuse)
I-690 to I-90 West (Syracuse to Buffalo)
I-90 to I-290 (Buffalo to Niagara Falls)

If you want to see Niagara Falls, get off at Robert Moses State Parkway (past the Grand Island Bridge). Take the parkway to the bridge to Canada. Take the QEW to Toronto

If you want to bypass Niagara Falls, take the I-290 to the end and enter the QEW to Toronto.
