Describe how the concentration of an unknown substance can be determined if a standard solution of protein is available?

Using the known protein solution, you can prepare a series of samples with different concentrations of protein by diluting the standard protein solution.

Then, you run the assay (biuret or Lowry or whatever test you're using) and generally you'll measure the color (absorption) of each of those know and unknown solutions.

Next you'll use that data to draw a graph that relates absorption to the concentrations of your known protein solutions. Finally, you'll take the absorption of your unknown solution and use the graph to determine the concentration of protein in that.
