You have some choises to pick up...
If you have a car or if you get a taxi, it is about 25 minutes! But I don't suggest you to get a taxi cause you are a tourist and may ask more money than the route costs to get to the port! The real amount that is usually asked is about 10 euros!
The other choise you have is to get the metro! The metro station is really close to Acropolis, the ticket costs less than 1 euro! You will be at the port in 35 minutes!
You have also the choise to get a bus- the price is about the same with the metro- but I will not reccomend it either because you may get lost in the center of the city!
If you need any further help I will be happy to inform you!As a travel agency I can give you any kind of information that you may need!

Get the metro at Piraeus to Monastiraki or Thiseio and walk a bit, or change at Omonoia, and then get the red line to Acropolis station. It should not take more than 45 minutes, say 1 hour with any delays you may have.
You could take a taxi, just make sure the driver doesn't swindle you (they try to get more money from tourists).

About 9 km of distance, but the time you need to go from Akropolis to Pireus depends on the traffic. Try the metro, it will take only 20 min.
