What is the meaning of the title of the book? does it have any symbolism behind it?

If it has any symbolism, I fear you have to find it for yourself. To me, it is quite clear.

The legend of the great hound killing Huge Baskerville in the 1600's provides the basis for Roger Baskerville's attempts on the lives of those who stand between him and inheriting the Baskerville estate.

The book is largely concerned with a ghost hound that "haunts" the Baskerville family. When it appears, someone in the family is supposed to die.

It refers to the hound that's though to curse the Baskerville family. The idea of it probably goes back to Hade's 3-headed hell-hound from ancient Greek mythology (I cannot remember the name of the dog to save me, though).

I doubt it...Doyle wasn't a writer who dealt much with symbolism and literary criticism. Reportedly, he got the idea from a local legend told to him by a friend he visited. I suspect the title is just a title.
