There are a few ways.

However, a common method for microorgs that are hardier than others is to use a Petri plate. The plate must have some sort of nutrient base in it on which the organisms can grow, such as agar or beef broth. Often, other nutrients and minerals must be added because the organism can not produce them on its own - nitrogen, sulfur, etc.

Be careful, though, because most species require a special blend if you want them to truly thrive.

After you get your plate, you take a purified sample of the species and spread it over the plate. You want to swipe in a zig-zag fashion, diluting your sample as you go, so that you can isolate a single colony. That's important because one colony represents one individual organism that has reproduced - so you get identical genes in that colony.

Incubation and time are required. Temps vary per species, as does the time, since reproduction rates vary a great deal.

There are several ways to culture microbes, just as there are different kinds of microbes to culture. Agar plates are useful for growing colonies from a swab or swipe, for identification or clonal selection, but liquid broth cultures are by far the most common method of culturing microbes. This can be anything from special broth to grow particular organisms to a simple hay infusion (dump some grass or hay into a beaker of water and let it sit for a while) to grow up all sorts of stuff that naturally occurs in the field.
