It's a gateway to the spirit world. If you enter it you can't return because you lose your body when you enter. Normally you would have to die to get to the spirit world, but this is another way in. It's in the department of mysteries because it's a mystery how it works and why it exists.

The thing with the department of mysteries is that they study the unknown. The Gate way is a gateway to the "other side." This is a bridge that can only be taken one way. Harry claims to hear whispers so I believe those "in tune" could actually understand what is being said. In this matter the wizards studying death believe they could learn more about death and the world that lay beyond it.

A gate way to the spirit world. Those who pass through it die. Sirius had just been struck by a killing curse from Bellatrix, had he not passed through it then there would have been remains to bury. Lupin seems to know about the curtained archway and what it is as he prevents Harry from following Sirius so as not to lose him too.

It could be a chamber like the ones at gringotts where u get trapped and are opened after a very long time. sirius could have died from hunger there.it could be to capture thiefs at the ministry.

another answer is that it could be some sort of gateway into non bieng, which is to say, everything..

From what I've read, and been told, and guessed, It's a passage to the realm of spirits (aka. the ethereal plane).
