Jimi Hendrix for his left handed 12 string (brilliant)
Stevie Ray Vaughn - responsible for the revival of blues
Eric Clapton
David Gilmore
Johnny Lang - child prodigy
I have to say this about Lang, he opened for Stevie Ray when he was still a babe. Anyone who has followed Lang's career knows he was a child prodigy who landed a major record deal by age 15. When kids his age were playing high school sports and anticipating the prom, the young blues phenomenon was touring with legends such as Aerosmith and B.B. King - not to mention headlining all over the world.
Many of the YA folks will be too young to remember Jimi Hendrix was the most influential guitarist in history. He was a self-taught guitarist, Hendrix played a Fender Stratocaster guitar turned upside down (so that the right-handed guitar could be played left-handed) and restrung to suit him. Hendrix pioneered the technique of guitar feedback with overdriven amplifiers, incorporating into his music what was previously an undesirable sound. He gained fame in England, and headlined Woodstock in 1969. He died in 1970 at the age of 27, the same year as Janice Joplin.
Eric Clapton and David Gilmore's works stand on their own, I think everyone knows them.
Stevie Ray, many of the young people may or may not know who he was. He was an astonishingly accomplished guitar playing, SRV ignited the blues revival of the '80s. He drew equally from bluesmen like Albert King Otis Rush and Muddy Waters and rock & roll players like Jimi Hendrix and Lonnie Mack as well as the stray jazz guitarist like Kenny Burrell developing a uniquely eclectic and fiery style that sounded like no other guitarist, regardless of genre. SRV bridged the gap between blues and rock like no other artist had since the late '60s. For the next seven years, Stevie Ray was the leading light in American blues consistently selling out concerts while his albums regularly went gold. His tragic death in 1990 only emphasized his influence in blues and American rock & roll.

To "Smart E"...it's Eddie VEDDER, and he,by no stretch of any means, is a great guitarist.
As for the top 5 I have personally seen: BUCKETHEAD, Yngwie Malmsteen, Joe Satriani / Eric Johnson, Dave Murray / Adrian Smith, B.B. King.

Well, since maze-wanderer beat me to it (he apparently wandered out of his maze before I got my head out of these contract reports), I'll just assume you meant gutarists and say:
1. Ace Frehley - I know he's not technically great, but his style has inspired countless other players
2. Dimebag - I saw Pantera open for Skid Row way back when. I never cared for Pantera, but I was impressed with Dimebag's playing.
3. Joe "F'n" Perry - The walking, talking picture of cool, and he's still one of the best guitarists around.
4. Mick Jones (Foreigner) - One of the most under rated players of his day.
5. Nuno Bettencourt (Extreme) - I never cared for Extreme, but damn could that dude play.
