You have to be extremely confident to have any success in acting at all.
Also, if your brother is a well-known professional actor, you certainly don't need to be in here asking this question!!!

Do you mean when you say the lines they sound lame? That's most likely because the words are unnatural to you. Maybe this person speaks in a different way from you, so it feels weird. Also, in some cases (more so in screenplays than plays) the script itself is just poorly written, so the lines are very ackward. Welcome to the world of acting!! An actors job is to bring a chracter to life that may speak and move very differently from them and make it seem completely realistic and natural.
So, how do you do this?
Think about the character, think about the characters objective in the scene and in individual lines, think about what the character is trying to get out of the other person in the scene. Characters must always have an objective, otherwise it would be boring and we don't want to watch. Think about what that is and how your character would achieve it. As for the actual delivery of the line, that will depend on what kind of person your character is and how they would go about achieving their objective. 1 line can be said many different ways, but whether you character is loud, shy, sexy, conniving, etc will determine how you deliver your line.
Also, for an exercise to do with your brother - determine what the objectives are in the scene for both characters. You each decide on your own for your particular part and you don't have to tell each other. Then, do an improv scene (as any character, you can make one up, just be yourself, whatever) trying to achieve your objective. This will help you understand the objective in the context of the scene better and will be easier since you can do it in your own language. Then, try the scripted scene again, still focusing on the objectives and see if it helps your lines sound more natural.
Break down the scence, just focus on a couple lines at a time.
If it still feels uncomfortable, sometimes just doing line reads in a bunch of different ways helps and one particular delivery just click and feel right.
It's hard to give an acting class in a short space, but hopefully that will help some.

ETA: Don't worry about being nervous! Professional actors get nervous before auditions all the time!! The more you do, the easier it gets to channel that nervousness into energy for your audition. The more prepared you are and the better you feel about your performance, the less nervous you will be and more excited to go and show them what you can do! So, rehearse, rehearse, rehearse!!!

If you are that nervous, you have no business auditioning for anything. Get some experience. If brother can't help you,nobody can.,
