How would I write that as an equation? I'm confused!

1 / 4^(-6) = 4096 is an equation.
4^6 = 4096 is also an equation.

(1/4)^ -6 = 4^6 = 16^3 = 256 * 16 = 4096

That is an equation, both side are true and equal:

(¼)^ -6 = 4096
(0.25)^ -6 = 4096

(4)^ 6 = 4096 to avoid a fraction and negative exponent (preferred)
