In ancient times it was the discovery, ability to generate and to use fire. next a little while later wa the invention of the wheel, followed by the written language, afte that you have to wait until the 1870s, then the harnassing fo use of electricity, when suddenly man made a huge jump into the modern world, things had not changed much for the everyday person in thousands of years, things where inproved on, yes therewas the printing press, but man already was writting, there was gun powder, and that was big, but did little to change ways of thinking and lifestyles in nations as a whole, also the steam engine was big, but they all pale in comparison to the use of electricity, it brought the electric light, mass communication, rapid communication, tv,radio,computers, appliances, hospital equipment, and the list is endless, take away all forms of electricity, and go back to a time before the 1870s , the entire earth leaped more ahead since the 1870s than the 10,000 or so years of known civilizations before.

The single most important achievement of course is developing a viable source of clean water and distributing that source in such a manner that is sanitary, free of contaminants. Any achievements like a pyramids is useless when the civilization is wiped out from the plague or disease or whatever due to poison or contaminated water supplies

in terms of the generic whole of mankind, i would say the ability to mass produce. starting with the cotton gin and its inter-replaceable aprts, through the assembly line of henry ford's model A (any color, so long as it was black), all of which made RECEIVING further improvements more affordable to the average human being. the computers we are all using at this very second would still be cost prohibitive if it weren't for the mass production methods seen today.
and it ranks highly because so many people on a worldwide scale have seen benefit from it compared to agricultural knowlegde (though a very good answer) or the printing press.

We are the first and only creatures that inhabit this planet that have the ability to mass adapt their terrain to suit their living needs versus having to accept what nature gives you.
Humans have the uncanny ability to adapt and overcome things that no other creature that inhabits the earth can do. Sure a cockroach will survive a nuke, but it cannot really do much productive in the world as humans can.
We learned to harness fire, something every creature was afraid of but something we needed for we were the only species without fur on us, so we had to clothe ourselves. Make our own tools and weapons over using our teeth and nails as an animal would.
that is what i think mankind's biggest acheivment is, the ability to adapt and overcome.

All the answers already given are good, but I would say the most important, life-altering achievement of humans has been learning how to farm. The move from pastoral/nomadic lifestyles to settled/sedentary lifestyles made all the other "achievements" possible.

writing Civilization is the accumulation of knowledge and writing allows the storage of knowledge so it is not lost. New technologies that increases the spread of knowledge like the printing press and the computer/Internet changes the world.
