It's a tossup between the Roman Empire and the British Empire. The Romans expanded their territory from modern day Italy to parts of North Africa, central Europe, the Middle East, and up to modern day Britain.
The British, on the other hand, expanded their influence into parts of Africa, Asia, and Canada for awhile.

The Empire of Charles V: Spain and its New World possesions primative though they may have been at the time, Austria, Bohemia, Sicily, Sardinia, Naples, Milan, and the Holy Roman Empire. Not to mention he was married to a princess of Portugal which gave his family blood connections to them as well.
