Let's see... possible interactions...

Audition (introduction, cold reading, readings with other actors, possibly some improv, trying multiple characterizations)

Callback (more of the above, usually with other actors)

Character Development (phone conversations, research trips, discussions, meetings, etc., costume/makeup approvals, etc.) - usually ongoing throughout the process, Director will brainstorm with actors to help them develop characters that fit the director's vision.

Script Read-Through (Table Read with all major actors)

Rehearsals (Either pre-filming or day of filming... usually start with the script and possibly move into improvisation)

Filming (Director will give feedback between takes, between shoot days, could allow for improvisation)

Re-Shoots (Depending on what's going on, the character may need to be changed or motivations different...more feedback)

Throughout the process, the director may also engage in social interaction with actors depending on the director and what kind of a relationship they're trying to establish with the actor. They may also encourage/discourage interaction among cast members. For example, it's usually easier for people to play lovers if they're comfortable with each other outside of the film environment than if they're complete strangers that have never met before.

I have never been asked to improvise at a movie audition, I have been asked to improvise my own dialogue in a scene, the only rehearsal we had was immediately before the takes, there were never any group read throughs, and once I went to the wrap party before we shot my scenes, because it was the only time the producer could schedule it, I only remember the producer being there, not the director.
There could be intereaction if the director is asking you about how you are going to play the character or the scene, or if you ask the director for help before the scene, such as having him check some looks you may be practicing for the scene.
