Because superglue requires air to trigger the chemical reaction. When it's sealed in the tube it doesn't have a chance to stick.
But that's much more dull than saying it's a consipracy by gluemakers and is actually the key to the universe!

Those answers that say superglue required air to make in stick are wrong. It is the opposite. Superglue will only work when all the air is excluded.So you put the glue on and press the two surfaces together to exclude the air and it sticks. When you close up the tube you make sure air is allowed to get back in to replace the glue squeezed out otherwise it will go hard in the tube.
