There is an old technique called a "stage kiss" which is a way to look like you are kissing when you are not. Ask your director to teach you how to do this, I bet your scene partner will be happy you did.

There are many things you can do.
1. The worst, but if you're completely scared of bombing, it's better than ruining the play--ask the kiss to be edited out or re-written.
2. Practice. If this is your best friend, make a joke out of it, be silly, and just have fun. Trust me, it won't be ALL that bad.
3. Do the stage kiss! Look it up online and then practice it with your friend, though if you have to "make out" kiss it may not work all that well.
4. Ask the director of the play if you can swap parts with someone else if the play is far enough behind for you both to learn your new lines.
5. Have fun no matter what road you take!

Honestly, I say sit down with your friend and your director and discuss it. If you are both uncomfortable, do the stage kiss, or edit it out or something. The problem is that unless the stage kiss is done really well, the audience will know it was fake, and you will loose a bit of the audience's respect. Kisses on stage are only that: staged kisses. They are as much part of the story as the charactor you play. No one expects you to really be that charactor in real life, and if you don't make a big deal about it, no one will think anything of your kiss either.
If you decide that you will actually kiss, and you are still nervous, talk to your friend and then practice. I'm sure they're just as nervous as you, and they will be relieved that you took the mature first step.

Honestly, the best advice I can give is to talk to the director. Many directors are willing to run a kiss scene at a rehearsal where only those involved in the kiss are there. Trust me when I say that this is one of the best ways to ease into something like this. Many directors even plan to do it on their own.
And I do have to say that stage kissing seems harder than it is. Luckily I've always been friends with the guys I've had to kiss and that does make it easier! Good luck!

Good advice from everyone, but no-one explained the "Stage Kiss".
Say You are the woman: Stand to the Left of the Man. He grabs you by the waist, and spins you so he faces Upstage. He then pulls you in so your faces are about 1" apart, and you are bent slightly backward, with his arm as the pivot point for your back. Count 1,2,3 ,4,5 and break, spinning back to your original position. You may, if required, collapse in to His arms, joyfully professing your Love and Devotion (or whatever is next in the script).
The whole idea is that your faces are covered from the audience. You do not actually need to lip-lock, just make it SEEM as if you are kissing. If, while doing this, you want to give a friendly little peck to say "Thanks for working with me", it is not beyond the bounds of propriety. Just don't crack up. It will ruin the scene ;).
I cannot tell you how many times I have had to "kiss" an actress I despised. My Character is in love with her Character, not I with her. This is why they call this "acting".
If you actually must do an on-stage, real, no-kidding kiss, pop a breath mint before the scene, commit to the scene, and just do it. It is just polite.

If you aren't comfortable doing it tell whoever is running the play.But whoever you're kissing is probably just as nervous as you,so I wouldn't worry about it.Plus do you want to ruin the whole play worrying about one kiss that doessn't even mean anything?
