The question of "how much do you want to spend? is relevant but I will make some suggestions for a modest budget. First get an ez-link card for the bus and trains. You will lose the deposit but the convenience is worth it and you do get a discounted fare so it may work out.
The Night Safari and Zoo are both worth while. If you have the stamina you can do it in one day.
The Bird Park is good but you must see the Nocturnal World or House of Darkness. It's enclosed natural environment where they've used artificial lighting to turn the clock around for the inhabitants.
Really worth seeing.
Go to Pulau Ubin. You can take a cheap ferry from Changi Point. No. 2 Bus is a good way to get there and you can drop off for a visit to the Changi Jail Museum on the way. Other wise you can take a bus from either Tampines or Pasir Ris MRT. Double Check that. On the Island you can rent a bicycle for about $4 dollars a day to ride around the island and see what old Singapore might have looked like.
I haven't been to Sentosa for years but I hear there's lots to do and see now.
For Shopping go to Jahor Baru. You can take the train to Woodlands MRT and Bus 2 bus services connect to take you to the Causeway. After clearing immigrations you will re-board the bus and go to JB Immigrations. From there you can take a taxi to many Shopping Malls where the shopping is much cheaper. Food is cheap there also and many Singaporeans make the trip just for a sea food dinner.
For a nice meal in Singapore go to the Polo Club. As a visitor, you can go in and have a nice meal in a very pleasant ambiance at a reasonable price. They take cash or credit cards. If you go on the week end you might see a match. BTW all clubs in Singapore are reasonably priced.
The Safari Yacht Club also has nice dining and you can hire a sail boat, with skipper if required, for an evening sail. You can do the same at Changi Sailing Club but it's not as nice although it's easier to get to. Number 29 Bus goes there from the Pasir Ris MRT. If you do the Jail Museum separate from Pulau Ubin, take the No 2 to the village then walk or take the 29 to the Sailing Club.
Hope this helps.

1) if u've a deep pocket & budget is no issue, u can consider shopping till u drop.
2) if u havent the luxury of budget, then i'd suggest u travel simple, savor all the local delicacies u can find.
3) if feasting is not what u do best, then sight seeing & admiring the light polluted nightscape by night.
4) if u really have nothing to do, find a sweet spot in a cafe or local coffee joint, take a round island ride on the MRT - blind yourself to the things u see, but listen intently - the ambient noise sgporeans create cannot be found elsewhere.
5) if u really really have nothing better to do, watch & appreciate retirees who're forced out of retirment (for the sake of survival), clearing dishes at eating places, sweeping & maintaining cleanliness... etc. offer them a drink or a meal.... u'd find that in doing so, could be more gratifying than anything else u do on this visit.
cheers!

I have no idea why so many people like to suggest for you to go to the zoo.. especially to see imported wildlife. The zoo is one of the popular attraction and its very well constructed. But it is hardly Singapore.
Singapore has been urbanized in many ways. Traces of the past are now barely visible, especially if you are not looking carefully. You may like to visit historical buildings and sites. Take a map and walk in the city. Starting from Victoria theatre, walk over to Fullerton Hotel (which is restored from the central post office of the colonial time) and on to boat quay & clark quay where commerce used to trive along the river for goods arriving from the sea. Go to Arab street to see how the old shop houses used to be, I think thats the last remaining block of shophouses that are not fully restored to a modern fascade.
Spend a day in Sentosa. Go through the defences of WW2. Then lunch along siloso beach, where there are a number of newly established pubs and restaurants.
Go to Chinatown to buy some small token of gift for your friends back home.
Go to Little India and Bugis for sights.
At night, there are lots of pubs and outlets to hang out at Mohd Sultan Road, Clark Quay and Emerald Hill.

Some of the suggestions above are worth considering. Just to add on...
If you love the sun and beaches, then it's a must to visit the beaches at Sentosa. Beach volleyball, swimming, suntanning are the few things you can do at the beaches. Then you may wanna go to the rest of attractions such as Skyride & Luge, Underwater world (foot reflexology is a must try!), Songs of the sea(only at night) etc. There are tons of beach bars too. And a night club called 'Cafe del' Mar'. I suggest you check out the website @ www.sentosa.com.sg
Shopping & dinner @ Vivo City. There are tons of restaurants there. [Harbourfront MRT station]
or shop along city area such as Orchard road, Suntec City & Marina Square + Esplanade[City Hall MRT station]
Night life -> plenty of bars and restaurants at both sides of Singapore river. [Clark Quay MRT station]
East Coast Beach - Visit their seafood restaurants there.
Changi village - Some authentic local food.
Serangoon Gardens Chomp Chomp - Seldom tourists know about this food village at Serangoon Gardens 'cos its located inside residential area but Chomp Chomp is popular among Singaporeans, especially with the residents at Serangoon Gardens. Business operates after 5pm and it usually gets really crowded in the weekends so try to get here by 6pm. Some of their must-try food includes Hokkien Prawn Mee, Fried carrot cake, chwee kwueh, satay bee hoon, soya bean dessert etc.
How to get there?: Taken MRT (NEL towards punggol) and alight at Serangoon station. Then come out from Exit C. Walk straight towards the bus stop and board bus no. 317. Inform the bus driver you're alighting at Serangoon Gardens and seek his advice on where to alight.
[Bugis MRT] - Bugis street (budget shopping)
Electronic gadgets - Funan Mall (Cityhall MRT) or Sim Lim Square (Bugis MRT or Little India MRT) --> Remember to bargain...
Other places of interests, Zoo+Night Safari(A must to go Night Safari!), walk around Chinatown, Little India (24hr Mustafa mall)
Fort canning park.
If you're a lady, you may wanna pamper yourself with Spa & massage or manicure/padicure. Do ask around on where is worth a visit....
