Are you sure you are looking at the right directory with the short cut? You may have ended up with My Pics in two places and the short cut is looking at the old one that you are not putting stuff into. Right click on the shortcut and select Properties. On the shortcut tab there is a line Target which has the complete path to the folder.
While that is open, use My Computer or what ever it takes to get to My Pics and see if the steps match the path. A common problem is that you have a My Pics inside My Pics so the path ends with .../My Pics/My Pics/ from copying directories, sometimes accidentally.

Plug it into iTunes & whilst it pops up there visit the pictures tab & there you would be waiting to choose which folder (on your (My) pictures folder) you will possibly be able to desire to sync to the iPod. Then click sync/save on with & it is it.
