The chief difference is that the Plymouth Rock colony was established by people who were seeking religious freedom from the crown, while the Jamestown, Virginia colony was established by a company with a royal charter and a mission to seek profit in the resources of the land.

The other chief difference is that the Plymouth Rock colony was founded because the Mayflower, intending to land at Jamestown, made landfall hundreds of miles to the north, and had to set up camp for the winter. They ended up staying in Massachusetts Bay. Originally, the Pilgrims were granted some land in Virginia and they agreed to settle it to contribute to the local economy.

The first Europeans to settle New England landed in present-day Massachusetts. These settlers were Pilgrims and Puritans from England seeking religious freedom.
the Pilgrims, established their settlement at Plymouth in 1620, and developed friendly relations with the native Wampanoag. Most early settlers came from within 60 miles of Haverhill, England. The Pilgrims were soon followed by Puritans who established the Massachusetts Bay Colony at present-day Boston. The Puritans, whose beliefs included exclusive understanding of the literal truth of the Bible, came to Massachusetts for religious freedom. Dissenters such as Anne Hutchinson, Roger Williams, and Thomas Hooker left Massachusetts because of the Puritan society's lack of religious tolerance. Williams founded the colony of Rhode Island, and Hooker founded Connecticut.

Religious intolerance back in merry old England (and the right to enforce their own brand of religious intolerance here) drove the Puritans to Massachusetts. Imperialism and Tobacco founded Virginia. Cheap Native American slave labor was also an incentive.

The Puritans were fleeing the persecution of the Anglicans who were in power. Those who settled Virginia were the aristocrats from England fleeing from the persecution of Oliver Cromwell, the Puritan dictator in England at the time. Many were also coming here because of the prime facie law of inheritance which gave everything to the eldest son upon the death of the father.
