Great question! Personally, I think there are many different ways to tell what kind of person someone is by conversing alone.

For one, pay attention to not just the voice of the person, but also their facial expressions, their tone, their small movements. Do they speak slowly, or quickly? Do they narrow their eyes? What kind of words do they use? Look at their body stance. Watch how they smile.

It's also important to watch them even when you're the one talking. Are they REALLY listening to you, or are they just nodding along? Do they make strong eye contact? Do their expressions mirror yours?

Just conversing with someone can let you learn a lot about that person. Among other things, you can tell if that person's lying, has something else on his/her mind, is shy and nervous, is trying to appear intelligent, etc. Pay closer attention and you can also find out deeper things, such as what kind of life that person has had, where he/she feels he/she stands in society, what he/she aspires (or doesn't aspire) to be, what mood the person's been in that day, and more.

Conversing helps us tremendously when it comes to analyzing, and helps us understand the other person better. We just need to remember to pay attention a little more.

The idea that a person can explain how to analyze a person's character, and do so by asking "how" on Y! Answers, is wrong. People are dynamic, for the most part. I can say "a person who does X Y Z during a conversation is a manipulative jerk who should't be trusted," but that's just one type of person. It takes experience in dealing with people and picking up on visual cues and use of language.
A good place to start would be to read a book about deception and how to tell whether or not somebody is telling the truth. That'll at least get you over the hump of knowing whether or not a person is honest.

By carefully choosing your topic of conversation. Throw some "off color" things in the mix and see how they respond.

by asking what they like to do and how they feel talking to you ask there hobbies and just think to your self is this a good person or not

How they act when you talk about certain things, how they react when they talk about certain things. How they take jokes. How they make jokes. What they talk about, when they're picking the topics.

It depends what you talk about. If they have a deviant charactor it will come out one way or another.
