Going through your files, changing your security settings, rewriting your harddrive. But then again, you can't really get a virus if you're not connected to the internet.

The dectection of the virus by an uptodate virus scan. If you don't have one, download AVG and move it to the "infected" PC and install it from a CD or other media and scan the system.

Other, less clear and positive (and basically useless) signs are a slow computer, strange locking of the computer and assorted things like that (but not BSoD, that is normally a driver issue). Unless you get a virus scan you can't be sure and you can't do anything about it expect reformat the drive and reinstall Windows.

If your not connected to the Internet then you probably don't have a virus, you might be looking at a problem with the operating system and just need to reload or at least run a repair.
