Yes. Both of Prince William's parents are descended from James I of England, who was the king when the colony of Jamestown was set up. On his father's side, William is James I's 12 x great grandson.

Yes. Jamestown was named after King James I (or VI in Scotland). James was William's great^12-grandfather. The full descent is:

King James I/VI
Elizabeth Stuart
Sophia of Hanover
King George I
King George II
Prince Frederick
King George III
Prince Edward
Queen Victoria
King Edward VII
King George V
King George VI
Queen Elizabeth II
Prince Charles
Prince William

I think some of the other answers got confused by the non-regal line of descent between James and George I.

In addition to descent through the Hanoverian line from his paternal grandmother, Queen Elizabeth, William is also a descendant of King James I of England (and VI of Scotland) on his mother's side, on the wrong side of the blanket:

Dianna Spencer (1961-1997)
Edward John Spencer (1924-1992)
Cynthia Elinor Hamilton (1897-1972)
James Albert Hamilton (1869-1953)
James Hamilton (1869-1913)
Louisa Jane Russell (1812-1905)
John Russell (176601839)
Elizabeth Keppel (1739-1768) *
Anne Lennox (1703-1789)
Charles Lennox (1671-1723)
Charles II Stuart's (1630-1685) liaison with Lucy Walters
Charles I Stuart (1600-1649)
James I (1566-1625)

Prince William and his dad, Prince Charles, are descended from a legitimate heir to James I (or VI), Elizabeth Stuart, and in turn through her daughter, Sofia, mother of George I (and the entire Hanoverian/ Saxe-Coberg/ Windsor line). Additionally, Dianna is a distant cousin of her husband through the Queen Mum.

------
*Elizabeth Keppel is an ancestor of you've guessed it, Camilla Parker Bowles, making Dianna and her successor descendants of the same king's mistress.

Jamestown, Virginia?

If so, yes, the Windsors are descended from the Hanoverians, who were descended from the Stuarts.
