Online scans can be a good test if you suspect your AV program isn't updating or scanning properly.
They can also be a nice check if the online scan is from a different vendor than your current AV. Products differ in how they identify threats.
The item to remember if you use an online scan is to temporarily suspend your current program so they don't compete with each other.

Because no virus scan program is perfect. They are created by humans, who can make mistakes in the coding, miss a virus, or not class something as a virus that another program would class as a virus.
Plus it is possible for a cirus to turn off you virus scanner so that (unknown to you) it is not working.
So checking your computer with a second, online, scan will assure you that your current scanner is working and caughting everything. And clean the computer if it is not.
But be warned that some (many) of the online scanners will find lots of 100's of viruses even though your PC is really clean in an effort to sell you their product. So check the reputation of an online scanner before panicing....
