For one if they try to avoid you and not make eye contact with you. Also when you speak to them and they give you one word answers.

When they try to steal your boyfriend/crush on purpose
When they are super mean
When they say they are mad at you
When they hate something and always say what they hate

if your friend is acting differently than usual, than you can pretty much decipher if they're still cool with you or not. Usually signs of neglect, carelessness signify anger or sadness towards another person.
