The "\SystemRoot\" directory is whatever directory you have the Windows program intself installed. On most computers that is C:\Windows. But if you have a computer with Windows NT, it will be C:\winnt. If you have a computer that multiple boots, it might be D:\Windows.
So the "\SystemRoot\" allows you to write a program and make it work no matter where the Windows program is installed.
On your PC, \SystemRoot\system32 is probably going to be "C:\Windows\system32"

That is a folder not a file.

There is no single file. The registry is stored in various places all over the HD. Some main keys locations are as follows. The rest are spread all over the place. Any .REG file on the disk is a potential registry entry. The following Registry files are stored in %SystemRoot%\System32\Config\: * Sam - HKEY_LOCAL_MACHINE\SAM * Security - HKEY_LOCAL_MACHINE\SECURITY * Software - HKEY_LOCAL_MACHINE\SOFTWARE * System - HKEY_LOCAL_MACHINE\SYSTEM * Default - HKEY_USERS\.DEFAULT * Userdiff
