How long do you have to seperate male female tiger barbs before you put them in the same tank to breed?

If they were nice, healthy fish before you separate them and it's only for final conditioning, a few days will be fine. Just be sure to really feed them well on live and froxen meaty foods before placing them all in the spawning tank.
