a mans man is not a gay term although it sounds it.... a mans man means that you are into hangin with the lads..not afraid to be masculine and dominant..... more Alpha male.... only fry your steak for 3 seconds and then fling your woman down on the bed kinda guy.......
a ladies man is kinda slimey....... cheesy white smile...... loadsa cheesy lines....... sell out your maleness cos its what you think women want to hear kinda thing......

I am neither but I will take a stab at the difference. A man's man is a guy who fishes, hikes, goes hunting and all those sorts of manly things including sports. They do not show pain or weakness..they are the kind of men that other men look up to and want to emulate.
Now a ladies man is not afraid to show his softer side. He can be a total dog but is not necessarily into sports, etc..he is into things that can get him women more than likely..I like a guy who is somewhere in between.

I'm definitely a man's man... but I am ONE ladies man
Most guys (if they know what one is) probably admire this type and want to be around this type....
as for the ladies men... they are also admired by most men .. the ones who are not jealous of them anyway... and certainly admired/wanted by woman, who prefer this type ...thats why women are never happy... women love confidence ... and ladies men usually are .... shy guys are not usually

Well, while I definatly prefer women to men, Im not sure I would classify myself as either.
Lets just say that Im a normal heterosexual male who's just trying to survive in this screwed up world of ours. As for the difference, I would hypothesise that it would depend both on society and the individual.
