Created in 1942 by Bob Clampett, Tweety's been a male bird since day one, even though he seems a bit on the "soft" side for a guy. If you don't believe me, you can check his biography right from his Looney Tunes website here:

Part of the reason he looks the way he does is because Bob Clampett based his design for the character on baby pictures of himself, as you can see in the comparative pictures here:

As you can also see, though, Tweety's undergone at least one important change. He began life in the pink... but that was changed in short order since the censors of the day complained that the bird looked "naked" when he was pink.
