It depends on the school. (Assuming that is what we are talking about.)
Running and standing tumbling
Jumps
Motion technique
Ability to pick up choreography
Ability to make up a cheer and/or dance
Willingness to do what you are told with a smile on your face
Spirit
Your physical condition
All of those things might show up on a judging sheet, plus even more. Ask the coach at your school to be sure.

I am a cheerleader for my high school varsity squad. For my schools tryouts we have to demonstrate a toe touch, a hurdler, and any double jump. We have to perform a cheer, a chant, and a dance that we learn at our clinics, which are for a few hours three days after school. Also, we can do an optional stunt such as a round off, back handspring, or any number of jumps. The judges score us on many qualities such as spirit, voice technique, and motions. for all-star, it's usually....
level 1- no experience required.
level 2- no tumbling experience, or minimal experience (cartwheel, roundoff...) and VERY RARELY a standing back handspring. no stunting experience, or minimal experience, maybe preps/elevators and a cradle. okay motions are looked for, although they will help you with that. they appreciate energy.
level 3- at a minimum, a standing handspring and a roundoff handspring. often standing multiple handsprings, maybe a roundoff tuck. be able to back/base/fly an extended one-leg stunt, a full down, and a basket toss with a skill. good motions should already be developed. high energy.
level 4- standing tuck, round off handspring tuck. rarely, a layout and add-on pass will be necessary. be able to back/base/fly a full down out of an extended one-leg stunt, a double down from either a prep or an extension, a basket toss with a skill (usually a kick full), and an advanced pyramid sequence. good motions should already be developed. high energy.
level 5- atleast a layout, and a standing tuck. really good teams will require a full and a specialty pass. be able to back/base/fly a double down from an extended one-leg stunt, a flipping pyramid, a basket toss with a kick full (a kick double will probably be required for the better teams), and a flipping pyramid. good motions should already be developed. extremely high energy.
this is not always what it is. but usually, it's along those lines.

well make sure you can at least do a backbend cart wheel and round off, splits too. You want to be able to catch on fast that way you won't have any trouble. You also might wanna be able to do a toe touch.

you need to do a toe touch, tuck jump, pike, hurdler, herkie.
you have to be flexible
you have to be strong
and you need to have personality and believe in yourself
