You can harvest tomatoes whenever they are the size and color that you are looking for. If you want ripe tomatoes, wait until they turn red, orange, or whatever color is right for the variety that you have planted. You can also pick them when they are green, and put them in a sunny window until they ripen.

Tomatoes are a tender perennial, so as long as the vines look healthy, keep them in your garden. If they happen to flower again, you will get another crop of tomatoes. But do protect them from cold weather, since below freezing temperatures will kill them.
