Several things are used for this purpose, but the most common is Sodium Thiopental, or more commonly known as sodium pentathol.
Sodium Pentothal is a yellow crystal, which can be dissolved in water or alcohol. It has alliaceous, garlic-like odor. If you were to drink a glass of water with Sodium Pentothal, it would depress your central nervous system, slow your heart rate and lower your blood pressure. All of the listed above affects are the functions of any sedative. Sodium Pentothal is commonly used as an anesthetic during surgery. The patient is unconscious within 30 to 60 seconds after the drug administration to the veins. The duration of the anesthesia is very short; it only causes a few minutes of sedation.

Police don't use drugs to get confessions, it is illegal. The only organizations that use this are in intelligence and the military, and even then it is used sparingly since it doesn't work that well on all people. It is mostly a myth created from the movies. The drugs used are available prescription drugs, primarily depressents and disassociatives that lower your resistance to interogation. They are fairly inneffective without considerable workup prior to their use. They are not drugs you can cook up in your basement, unless you are running meth lab and have access to the components. If you are thinking you can use this on someone to get information out of them, it won't work and most likely you'll be the one going to prison.

In the US, this would be illegal as it violates a person's right to not to self incriminate, the 5th Amendment. This confession would not be admissible in a court of law. Any competent attorney would get this throw out.
What the feds / spooks do, is another issue.
