The toilet in my spare bathroom is not working nor is there any water. The water was working just fine a few weeks ago and the valve is still turned on but no water is coming out nor do I hear the sound of water running.

I turned the valve off and on again and still don't hear or see water coming into the tank.

The water in the bathroom sink is working so I'm not sure why the toilet would all the sudden stop functioning.

I have well water, could the minerals in the water have caused a blockage in the lines or is there something else that would cause the water to just stop running? This bathroom is not used very often so the toilet does not get flused daily like the other bathroom in the house. Do I need to get a hold of a plumber ASAP or is this something common that happens.

Another thing to consider is that some new toilet fill systems have a 'leak protection' device installed. I just installed a couple of these the other day, and basically what it does is lock the float in the up position so that if you have a leak your water won't be constantly running.
If you have this kind of system, you won't have a float that looks like a ball on the end of a rod, but rather a large cylinder that goes around the tube where the water comes into the toilet. This kind of fill system will typically also have 2 chains going to the flush lever, one going to the flush flap itself and another going to a small piece of plastic under the float.
If this is the kind of system you have, the problem could lie with the small piece of plastic under the float. It is only about an inch by an inch and is hinged under the float. it has a small piece of metal that makes it lock the float in the up position if it's chain is hooked up correctly. You can wiggle around the parts in the tank around that area with the water turned on to see if it causes water to flow. If you get the water to flow this way, the chain going to the lock could be broken or loose, the float won't go down and you won't get any more water coming into your tank. To fix it, you simply have to readjust the chain so that every time you flush the handle, it pulls the lock out too.

Have you taken the lid off the tank to see if the float on the fill valve is stuck? If you have well water it would be best to put a plastic fluidmaster valve in. There are no metal parts to rust and the float will work much better. Most of the guys I know can change one, it takes 5 minutes and is really easy. They cost 10 or 15 bucks at wally world and come with instructions if you are brave enough to try yourself. The biggest problem with changing one is getting the reservoir tank empty so no water runs out on the floor. Then it's nothing but a pair of pliers (and maybe a wrench) to take off the water line (turned off of course) and unscrew a plastic nut on the underside of the tank. Nothing to it!

Turn off the water supply valve and disconnect the line that goes to it. (if there is water in the tank, it will drain out so have a container ready) Hold a rag over the valve and turn it on and off 2 or 3 times. This should clear any deposits in the valve and let you know if you have water to the valve. If you do have water, the problem is probably the fill-valve. This can easily be replaced with a Fluidmaster 400A fill-valve for about $10.

Sounds like the feeder valve to that toilet is closed, not the valve under the tank.

Do you have a leak elsewhere?
