It may be different across different states. Here in California, you're expected to:

-Know the hand signals (Left, right, brake/slow down)
-Know where the emergency light and defroster are located
-Check your mirrors
-Make safe lane changes (signal, look into mirror AND over shoulder at blind spot)
-Maintain speed limit
-Make safe turns
-Park next to a curb and go reverse about 3 car lengths
-Have your eyes constantly moving, don't develop a fixed stare.
-At stop signs, stop for a full three seconds before the white line for pedestrians. Then slowly advance and stop again for traffic, then proceed when safe.

They may or may not test you on parallel parking, depending on what state you're in. They didn't test me on it, but it's still useful to know.

Check your state's DMV driving manual for more information.

They will test you on your ability to...

1. Stay between the lines
2. Drive at the speed limit (not above or below)
3. Signal several seconds in advance of doing anything
4. Regularly look at your mirrors
5. Know who has right of way in traffic
6. Know which lane to turn into when making a turn
7. Maneuver your car in reverse

1. Look for stop signs. (On my very first test, there was a stop sign (not on a pole, but attached to the side of a pillar somewhere, and I didn't see it, as it was not noticeable. Needless to say, automatic fail. Later, the person who drove me to the test asked me about why I didn't pass, and I told her, I missed the first stop sign in the parking lot. She said, "what sign"? I said, "the one back there, you just ran it" LOL, don't put stop signs up against red brick backgrounds and expect people to see them! ... it wasn't even an intersection, it was just a "gotcha" to me.) 2. Look (turn head) before changing lanes. (I checked the mirrors, but I didn't look all the way around prior to changing lanes. This is potentially dangerous! This was a violation, but I don't think I failed for it, it was the stop sign.) 3. Yellow lights mean try to stop, not try to run the light. (I know people may know better. Yellow usually means stop, if safe to do so, not ... how can I beat this red light? I tried to beat the yellow light, and ended up running a red light... Of course, another auto-fail on the same test.) 4. On my second test, I was accused of turning too wide. (So, tighten up your turns.) 5. On my third test, I passed. You should do better than me. My third test was my third time driving, LOL.
