Indeed, as others have pointed out, it can mean both. However, I would imagine that 99.9% of the time those words are uttered, the purpose is 'allow us to play'. If it were the former, the person speaking would most likely say, "Let's play."

both are correct when said in different contexts.
for example , if your mother does not allow you and your friends to play , you may request her in apleasing manner,"mom, please let us play"
and when you call your friends for a game you say"come,let us play".
