If you're into Googling, I found this. I would vote for Janis because she was an entity onto herself. Grace Slick is more remembered for being lead singer for Jefferson Airplane.

I would think that Janis Joplin would have to be one of the first, however some people would call certain groups in the 50's rock and roll, and there were a lot of girl bands at that time.

I would consider Janis Joplin the first. Grace Slick comes to mind as well, but I would have to say Janis (and I was alive at the time).
