If you open your system case up, you can disable the wire leading to the power switch.
If your case allows you to lock it then you can disable the key press power switch and lock the cash shut.
It can then not be powered on without opening the case and hooking up the power switch.
Alternately, you can take out the RAM. This won't stop it from being turned on, but it simply won't work at that point.
Another trick is to simply pull the power cord off the motherboard that connects it to the power supply. There is usually a small white clip or hasp that keeps it on there, but once you figure it out its pretty easy. It won't turn on at this point.
It's often easy to trick people into thinking a computer won't work by changing the boot sequence in the BIOS. Make it boot off of things that aren't there (e.g. a removable drive, or network boot), you can leave a blank floppy in the drive, or a music CD in the CD drive, and it will fail saying "can't boot, no OS". You can simply flip the BIOS so that it doesn't use anything to boot. You can select the BIOS to use the wrong configuration for your hard drive and it won't boot.

Most computer BIOS have an optional system password which, if set, will prevent the computer from booting unless it's entered. This won't stop a potential intruder or thief from powering up, but it will stop them from loading the OS, even from CD or floppy.
Anyone who can open the computer's case can remove the jumper that will reset the password, so if you set a system password, you'll need to physically lock the case to make it stick. Most cases have a loop for a small padlock or other security device.

You may be able to in the BIOS.When the computer starts and before windows loads hit the delete key or whatever your brand says to get into Bios settings. Once in there look around for the wake function (if you have it).Set the time you want, save the setting and (F10 usually) thats it. Some computers will allow it to wake from a lan signal or moving the mouse or SET a time that you want to boot up at. Remember to set it for a time you will be around. If you want to start it before that time you will have to open the tower up and reset the BIOS (pull the battery out for two or three minutes and replace or clear the jumper.

I don't think that is possible, however, you can perhaps set a system password (done within the BIOS) as well as a Windows password; it will ask the user to enter a password before it shows the boot screen logo.

instead of using the windows security most computers come with thier own security, from the BIOS you can set a password for your computer. the password is requesed for even before windows starts, if you dont want your computer to come on well i think you have to disconnect your system fromm all source of electricity.

Yes, hiding the power cord and locking it in a safe, or opening the computer and disconnecting the power cables (DO NOT TRY IF YOU DO NOT KNOW WHAT YOU'RE DOING, YOU MIGHT POSSIBLY KILL YOURSELF OR YOUR COMPUTER!!!!!!!!!) I think I've heard about something that can do that with a key lock, but I think it's just a rumor (or, someone who has too much free time on his hands.)

Take out the cord? Flick the power switch at the back of the hard-drive?
I don't think you can block someone from actually turning on your computer.
