I was having the same problem with my new Sony Vaio.
You can try various things (see below) I had to go through all but the last of these steps myself.

1. Reinstall your updates - sometimes updates don't get installes completely. You can go to the control panel and sound devices for this.

2. Reinstall your drivers - Somethng may be wrong with your sound driver. If it is, this should help, but if there is nothing wrong with your driver, this won't do anything.

3. Check your sound service and make sure your sound service hasn't been interrupted. You can have your driver correctly installed and working properly, but if your sound service (what acutally reads your card) is suspended , or stopped, it will not work. Make sure it is active, or on (or whatever phrase is closest in your drop down menu). This isn't some service you susbscribe to, it's just like turning on your screen saver (internal).Use your control panel and sound devices. Fiddle around with the tabs until you see a "sound service" option.

4. Restore your computer to a previous time (when it was working properly). - When all else fails make you computer revert back to a point in time when it wasn't acting all buggy. Use control panel and restore.

5. Replace your motherboard - the sound card in most laptops are soddered onto the motherboard. I'm not sure if you can replace just the card in a tower, but i'm sure it is easy to find online. This may require a professional (unles you know what you're doing) because if you misplace, bend, or disconnect anything, your computer won't work properly.

6. When all else fails - sell it on ebay and buy a new one.

I had the same problem once or twice with vista. When doing updates, vista will periodically fail on an update. If this happens with the driver for the sound card, it's like having your speakers uninstalled. Try the update again and if it's sucessfull, it should re-install your speakers. If not, try going back to a restore point, where the old driver was intact. If that doesnt work, your gonna have to shell out to the geek squad...
good luck.

open your audio setting and make sure none of your settings are mute, some mallware will do that.
