No, they do not have 32 bit and 64 bit on the same disc. The OEMs come as either 32 bit or 64 bit. The retail versions come with the 32 bit disc, if you want the 64 bit disc you have to send away for it and Microsoft will mail it to you, for a fee. This does not apply to retail versions of Vista Ultimate, which comes with both a 32 bit and 64 bit disc. The Core 2 Duo is a 64 bit processor, and normal 32 bit programs can run in a 64 bit OS.

i. most oem packs of vista have the option during installation > 32 bit or 64 bit
ii. core 2 duo > is a 64 bit processor
iii. most modern computers with the core 2 duo presently come pre-installed with vista 32 bit

yes its on the same disk, but you have to buy the 64-bit version to use it.
