No such thing as one best. There are many for different reasons, the ones who had impact and made you play guitar...depending in which era you grew up in...
Eric Clapton, Jimmy Page, Jimmy Hendrix, Jeff Beck , Tony Iommi impacted the 60s. 70s and 80s. Space Ace Frehley simply made you play air guitar!
Eddie Van Halen, Steve Vai, Randy Rhoads, Zakk Wylde and Joe Satriani impacted the 80s and 90s. Dimebag Darrel, Nuno Bettencourt and the list goes on. It depends who made you pick up the guitar or rock out
a bunch that deserve this distinction
how about double guitar teams?
KK Downing and Glenn Tipton - Judas Priest
Dave Murray and Adrian Smith - Iron Maiden
Phil Collen and Steve Clark - Def Leppard
Jason Becker and Marty Friedman
Rock on brother!!

Sure Eric clapton, and Jimmy Page were the best heavy blues rock guitarist. Jimmi Hendrix the best pscyedelic guitarist, but did you know that Jimi loved ZZ tops texas bluses rock guitarist, "Billy Gibbons"? That say something if your admired by one one the greatest guitarist ever.. I also like another texas blues rocker, "Stevie ray Vaughn and also Steve Miller, Eddie Van Halen and Pete townshend and Jeff beck!

Impossible question to answer, but for the sake of argument in my opinion it'd be Jimmy Page because of how good he was combined with how much I like Zeppelin's music. Hendrix, Clapton, and Brian May are way up there on my list too.

We cant really answer this question because you have to break it down by genre. Dimebag would be my choice for best metal guitar but I would have to pick BB King over him if we were talking about blues. And dont forget David Gillmore if it was classic rock or Hendrix if it were trippy post hippie music. I can go on and on
