Sounds like the linkage on the latch has come off. It's a simple repair,,,,,,,,once you get the interior panel off. That's the real pain in the *** for you. Fixing the latch is easy, but trying to get that interior panel off without damaging it will tend to make even the most patient person cuss. It could definitely be a DIY fix, if you have the patience for it. If not, I recommend taking to a mechanic. It shouldn't take more than about 30 minutes to fix for a mechanic that knows what he's doing. Hope this helps.

Do you mean that the key will not go into the lock, or that it will not turn, or that after turning the latch doesn't release?
If the first or second, I would try some WD-40 to unfreeze the lock. If that works, wait about 3-4 days then sqirt some dry graphite lubricant into the lock.
If the third, can you open the door from the inside? You may have to remove an interior panel to get to the latch and spring it open (carefully, don't damage it). You may have to apply a little WD-40 to unfreeze it. Check the mechanism to see if anything has slipped or come loose. If so, adjust it. Clean any grime off the latch and apply a small amount of grease (white moly) on the moving parts.

PULL HARD!!
Maybe it's just jammed with some trash, clothes, or a pepsi bottle.
ADDED:
If that doesn't work, then take it to a body shop or something.
