The principal of falsifiability. A particular observation makes certain hypotheses no longer viable. For example, since enzymatic activity is intact after cell lysis, any ideas about a magical life force are completely invalidated.

All experiments are setup this way. If X is true then Y must occur. X is your hypothesis, Y is the expected outcome. It doesn't prove anything, but it strengthens the hypothesis if Y occurs as the result. (this is technically incorrect but I don't feel like discussing the concepts of null hypotheses, normal distributions and confidence limits. A quick wikipedia will probably let you know about these things. They are all very important in the interpretation of data.
