The Chicago Colts set the record for most runs in a game on June 29, 1897 when they scored 36 against the Louisville Colonels.
Since 1900, two teams hold the record. The Boston Red Sox scored 29 against the St. Louis Browns on June 8, 1950. The Chicago White Sox tied the record five years later when they defeated the Kansas City Athletics 29-6 on April 23, 1955.
The way runs are being scored today, we might not have to wait very long to see this record broken.

Answer
The MODERN record for most runs by a single team is 29, by the Red Sox against the St. Louis Browns on 6/8/1950, and by the White Sox against Kansas City on 4/23/1955.
Earlier,on 6/29/1897, Chicago of the NL scored 36 runs against Louisville.
The AMERICAN League record for BOTH TEAMS is 36 (Boston 22, Philadelphia 14; 6/29/1950)
The MAJOR League record for BOTH teams is 49 (Cubs 26, Phillies 23; 8/25/1922).
Added Note: The 1950 Red Sox were a potent team that year. On June 7, 1950 -- just one day before setting the single-team record of 29 runs noted above -- they scored 20 runs against the hapless St. Louis Browns. Added to their 22 runs on 6/29 makes THREE TIMES that year that the Sox scored 20+ runs in a game.
