The mix of R&B, the blues and country led to Rock and Roll. Elvis set this in motion with "That's All Right". This song "exploded on the world scene," and that's what made Elvis The King of Rock and Roll. All other artists before him didn't project their music internationally.
Check out this article from the Seattle Times in 2004. It pretty much tells you that no one artist can really lay claim to the first Rock and Roll Record.

I don't think it's possible to pinpoint what song was the FIRST Rock And Roll song. 'Rock Around The Clock' was the first Rock and Roll song to make to the Billboard charts but I think most of us know that Rock and Roll was around a long time before that. They just called it Rhythm and Blues.

This is one of those questions that doesn't have a "right" answer. Rock and roll has its roots in music that predates the phrase "rock and roll" by several decades. Surprisingly, the wikipedia has a good article on the topic. The url is:

The first number one song labelled as "rock and roll" was Bill Haley's "Rock Around the Clock" in 1954.
