to say 'I am not interested.' is a final way of ending the conversation in that you are not interested in anything. To say 'I am not interested in... implies that you may be interested in something else. It would be possible to add 'but' at the end and include something you are interested in. So to say 'I am not interested in taking a holiday to Mexico but I may be interested in taking a holiday to England.' is to add an alternative.

With telemarketers, the best way to say you're not interested is to say "One moment...let me get a pen." Then just lay the phone down and go about your business. A few hours later hang the phone up. :-)
