By hand- vertically, one hand high, one low, balance carefully.
By vehicle-you need a ladder rack. Or feet down in the extreme rear of truck bed and plenty of padding for where it rests on the roof. Tie securely.

You might want to hang red cloth from the ends of the ladder to warn other drivers of the overhang.
