Biggest one I know of currently manufactured is the KTM 690 Supermoto. Largest ever was probably the Suzuki DR800.

Well, there was the Suzuki DR800, but I don't know if it was mass-marketed. I know it was sold in Europe. Much like the DR650, it was a dual-sport bike. It was also called the Dr. Big.
Maico had a one-off 760cc two-stroke single (not technically a thumper, as it was a two-stroke). The editor from Dirt Bike magazine rode it in the Black Woods 100 enduro.
There are lots of singles in the 600-700 cc range (Suzuki, Kawasaki, Honda, Husqvarna, BMW, KTM, Husaberg).

I had a BSA 600 single,flathead. If I remember it was a MS 21. Very rare, I think. I would say the Yamaha is probably the most popular. I know Suzuki Has offered some large singles, but have no idea the size

Back in the good old days, single cylinder british bikes like the BSA 441 Victor or the Triumph 500 were the hot items...
Currently, there are several 650cc singles on the market from Suzuki, Kawasaki, BMW and others...
I can't think of any singles bigger than 650cc.

When I was young I rode a 500 Yamaha TT. I don't remember the thumpers having a large following but the did have some very avid fans if only a few.

Yeah, I believe it is the 650cc Suzuki Savage, now called the S40.

honda xr650L was the biggest I have seen, also suzuki savage 650
