Personally, I'd say none of these words fit me.
As others have pointed out these words each have their own levels of baggage to carry with them, be it of a religious nature or of a more general moral one...
"Purpose" is the word I use. Yes, it does sort of combine both the journey and the destination in one word here, but it really does the trick for me, in terms of answering a lot of the questions. Things like: Why am I here? or Why do I bother? Not to mention, in my mind, the word "Purpose" carries much the same sort of moral weight as more religious words like "Mission" or "Calling" but without necessarily having to *sell your soul* (such as it is) to get there. I can have a purpose, or more precisely choose a purpose freely of my own free will, without having to *disregard* my own intelligence and free will and *throw myself away* on the rocks of religion.
"Purpose" has all of the moral weight to me and little to none of the Bible-Burnt Baggage. ^_^

To each their own, in my opinion ambition is the most
inspirational. Ambition is driven by enthusiasm, and
enthusiasm is the most naturally sustainable inspiration.
Inspire by truly wanting to inspire; by pursuit of truth.
Motivation is best accommodated from within the
self's enthusiasm. When ambition is in alignment with
consideration, all of the other words are most efficiently
and thoughtfully carried out. When my mission is your
mission we both win. When my calling and your calling
are mutual then we can hear each other. When I know
my duty and you know your duty we can both confidently
work together. When our goal is progress then we can
proceed separately. When we are proactive towards
our aspiration then our aspiration is justified.

I would say CALLING, because it is the least pretentious it doesn't involve ruthless ladder-climbing and striving, like ambition. A duty is something you feel you must do, regardless of how inspired you may be. A goal is something worthy and (hopefully) self-generated, but hardly noble or lofty. Inspiration implies that it is lofty. Mission smacks of religious fundamentalism and colonialism. A calling does not have to be a religious calling.

Others views are bound to differ but for me the word "Duty" gives me the greatest source of inspiration..
I am very committed to my family, friends and country and almost everything I I live for is for them!
I feel its my "duty" as a daughter, sister, friend, granddaughter, neice, cousin, auntie and English citizen...
As I've said before..We are like passengers on a train..We get off at different destinations..One day I'm going to get off of that train..But before doing so, I want to make sure that I've done my DUTY!

How bout Serendipity, or Epiphany. Inspiration literaly means to draw in air. I would say that the two words I listed are more like a breath of fresh air, and there's nothing like a full tank to get me where I'm going!

I think that to get to my Destination, it depends on what I believe above all you mention is the right one for me, so to me it is Duty, because it was, and still is my Duty to take care of my Husband, till Death us do part.

I would say none of the words . Inspiration means to influence someone to do something that they had doubts if they could do it. It is the emotional aspect of influencing people. For example, a politician will influence you with his inspirations (even though they may be lying) and get you to vote for him. It motivates you amd gets you going. To inspire is to move one.
