You can look at them as fractions and say
2/78 ==> two seventy-eighths
5/65 ==> five sixty-fifths

or view them as divisions,

2/78 ==> 2 divided by 78 is
5/65 ==> 5 divided by 65 is

40:5 is pretty much considered an archaic form. I learned it in terms of ratios and proportions as "40 is to 5". For example:
40:5 :: 8:1 reads 40 is to 5 as 8 is to 1
In modern usage, it becomes
40/5 = 8/1

2/78 = 1 over 39 OR 1 divided by 39.

5/65 = 1 over 13 OR 1 divided by 13.

Both need to be reduced first.

For me, i've always said Numerator over Denominator, so 2/78 would be said for me as 2 over 78, not that i've paid to much attention to how i pronounce my maths, its the reason i'm not doing A-Level English :D
