As someone else said it on the Prinsengracht.

From Dam Square go around the palace and follow the street with the palace at your back until you get to the Westerkerk (Western Church). There's a nice sculpture of Anne Frank next to the church. Turn right at the canal (same side as Westerkerk). The Anne Frank house is in the next block. There are often fairly long lines, I've had good luck in the morning though.

Anne Frank's House is near the West Church (Westerkerk) in amsterdam's center. It is now set up like a museum. Worth the visit.Address is Prinsengracht 267, near the corner of Rosenstraat

It's just off the downtown area of Amsterdam and is open to the public.
